Course Title LABORATORY OF BIOCOMPATIBILITY AND CELL CULTURE

Course Description Course Aims The Biocompatibility and cell culture Laboratory aims at (1) describing the current regulations on biological safety; (2) presenting the techniques and assays for cell culturing; (3) discussing protocols and assays for the in vitro biological validation of biomedical materials and devices. Lessons The lessons will be focused on the following topics: (a) current regulations related to the classification of the biological laboratories and biological agents, risk assessment; safety rules and good laboratory practice; (b) biological environment-material interaction; (c) cell culture techniques, in particular referred to culture media, cell models, techniques, equipment and assays for cell culturing and cell response assessment; (d) in vitro biological validation of materials and biomedical devices: models and assays; (e) overview on in vivo biological validation of materials and biomedical devices; (f) design and execution of experimental protocols. Laboratory activity The laboratory activities will be assigned to groups of 4/5 students and will be focused on: (a) cell culture techniques: defrosting, expansion, cryopreservation; (b) cell seeding and culturing on polymeric/metallic/ceramic materials; (c) samples treatment and in vitro cell response assessment. Other activities To complement the described activities and to stimulate active learning, the structuring skills of thinking and knowledge and the assessment among colleagues, each group of students will be asked to prepare a selected "case study" assigned by the lecturer, related to the course topics, that will be discussed with the other classmates in flipped classroom modality and also with external lecturers/scientists. Expected acquired knowledge At the end of the course, students are expected to: (a) have a good knowledge about the theory and the practical aspects to design and discuss an in vitro validation protocol for biomedical devices; (b) have acquired the basic knowledge of the in vivo biological assays; (c) critically discuss the methods and results of in vitro biocompatibility assays described in literature. Exam The exam has a written and an oral part: (a) the written part of the exam requires the production of a report based on the experimental activities performed in the laboratory activities; (2) the oral part requires the presentation and the discussion with the lecturer of the previously mentioned report content and an in depth discussion to evaluate the topics treated during the lessons. Further information To attend the course, a basic knowledge of cell biology is suggested. The language of the course is Italian. The attendance of the lessons and laboratory actitivies is mandatory, In particular, the participation to the lessons focused on biological risks and chemical risks is mandatory to access to the laboratory activities. The teaching materials will be provided by the lecturer during the course through the related BEEP portal.
